Overview

STIL-101 is an autologous tumor-infiltrating lymphocyte (TIL) therapy developed by City of Hope for the treatment of advanced solid tumors. The manufacturing process leverages the **PepSeq** technology, a proteomics platform that screens DNA-encoded peptide libraries against a patient's HLA class-II proteins, combined with in-silico HLA class-I binding models. This approach identifies and prioritizes potential neoantigens to selectively expand neoantigen-reactive TILs, aiming to overcome the limitations of traditional TIL expansion methods that often result in T cell exhaustion and lack of selectivity. STIL-101 is administered as a single infusion following nonmyeloablative lymphodepleting chemotherapy and is followed by interleukin-2 (aldesleukin) support to promote T cell persistence and anti-tumor activity.
