Stilbene 5c, chemically known as cis-3,4',5-trimethoxy-3'-aminostilbene, is a synthetic stilbene derivative that acts as a potent microtubule-disrupting agent. It exerts its antitumor activity by binding to the colchicine-binding pocket of tubulin, which inhibits microtubule polymerization and leads to cell cycle arrest at the G1 and G2/M phases. Preclinical studies, primarily conducted at Virginia Commonwealth University, have shown that stilbene 5c induces multiple pathways of cell death, including apoptosis, autophagy, and mitotic catastrophe, as well as promoting senescence. It has demonstrated efficacy in various cancer models, including ovarian cancer, leukemia, HCT-116 colon cancer, and B16F10 melanoma, notably functioning in a p53-independent manner.
