STK-009 is a polyethylene glycol (PEG)-conjugated mutein form of the cytokine interleukin-2 (IL-2), specifically engineered to selectively activate CAR T cells that express an orthogonal IL-2 receptor beta subunit (hoRβ). It is designed to enhance the proliferation, survival, and anti-tumor activity of these engineered CAR T cells while avoiding stimulation of normal lymphocytes such as NK cells and native T cells. STK-009 is administered as part of a two-component cell therapy regimen with SYNCAR-001—a CD19-targeting autologous CAR-T cell product expressing the engineered hoRβ receptor—enabling precise control over CAR-T expansion in vivo. The combination therapy is being developed by Synthekine for indications including systemic lupus erythematosus (SLE), lupus nephritis (LN), and various CD19-positive hematologic malignancies[1][3][5][6].
