Overview

STM3675 is a preclinical, small-molecule inhibitor of the RNA methyltransferase METTL3 (methyltransferase 3, N6-adenosine-methyltransferase complex catalytic subunit) being developed by STORM Therapeutics as part of its RNA-modifying enzyme pipeline for oncology. By blocking METTL3-dependent N6-methyladenosine (m6A) deposition on RNA, STM3675 reduces m6A levels, induces apoptosis, and impairs proliferation in acute megakaryoblastic leukemia models driven by the RBM15-MKL1 fusion oncoprotein, while modulating oncogenic pathways such as Wnt signaling and expression of Frizzled receptors.[1][5][9][12] Preclinical data indicate that selective METTL3 inhibition with STM3675 disrupts leukemogenic m6A programs and R-loop homeostasis, supporting further development of METTL3 inhibitors as novel targeted and immunomodulatory therapies in hematologic malignancies.[1][5][7][11][12]
