STM9201 is a small molecule inhibitor of METTL3 (Methyltransferase Like 3), a key enzyme in the epitranscriptomic regulation of RNA through N6-methyladenosine (m6A) modification. Developed by Storm Therapeutics, STM9201 is being investigated for its potential in treating medulloblastoma (MB), a common malignant pediatric brain tumor. The drug works by reducing global mRNA m6A levels, which in turn decreases cell viability and suppresses tumor growth. Research indicates that METTL3 inhibition by STM9201 can also enhance the immunogenicity of tumors by modulating pathways related to the innate immune response and cholesterol biosynthesis, potentially overcoming immune evasion in aggressive brain cancers.
