Overview

STN1014100 (also known as AR-15512) is a first-in-class, potent, and selective small molecule agonist of the Transient Receptor Potential Melastatin 8 (TRPM8) channel. Developed by Santen Pharmaceutical under license from Aerie Pharmaceuticals (now Alcon), it is being investigated as an ophthalmic solution for the treatment of dry eye disease (DED). TRPM8 is a cold-sensing receptor located on the corneal nerve endings; its activation is thought to stimulate natural tear production and provide a cooling sensation, thereby improving both the signs and symptoms of dry eye. In clinical trials, STN1014100 has demonstrated the ability to increase tear film stability and reduce ocular discomfort. The molecule was originally discovered by Avizorex Pharma before being acquired by Aerie Pharmaceuticals.
