STP1 (formerly STAC-102) is a first-in-class precision medicine candidate developed by Stalicla for the treatment of Autism Spectrum Disorder (ASD), specifically for a subgroup of patients identified as ASD Phenotype 1 (ASD-P1). It is an oral fixed-dose combination of two small molecules: arbaclofen, a GABA-B receptor agonist, and bumetanide, an NKCC1 chloride transporter inhibitor. The drug's mechanism of action is designed to restore the excitatory/inhibitory (E/I) balance in the central nervous system, which is hypothesized to be dysregulated in certain neurodevelopmental disorders. Arbaclofen acts to reduce excessive glutamate release, while bumetanide lowers intracellular chloride levels to enhance GABAergic inhibition. STP1 was identified using Stalicla's proprietary DEPI (Data-driven Enrichment of Patient Identification) platform, which matches specific drug profiles to patient subgroups based on transcriptomic and genetic data.
