STR-P005 is an investigational mRNA-transfected, CD19-targeting chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of relapsed or refractory autoimmune diseases. Unlike traditional CAR-T therapies that utilize viral vectors for permanent genetic modification, STR-P005 employs mRNA electroporation to achieve transient expression of the CAR on the surface of T cells. This transient approach is designed to provide a controlled window of B-cell depletion, potentially reducing the risk of long-term B-cell aplasia and associated infections, which is a significant safety consideration in non-oncological indications. Due to the temporary nature of the CAR expression, the therapy is typically administered in multiple infusions (e.g., every 3 or 4 days) to maintain therapeutic efficacy. STR-P005 is currently being evaluated in early-phase clinical trials, including investigator-initiated studies at Beijing GoBroad Hospital, for various B-cell mediated autoimmune conditions.
