Overview

Streptococcus pneumoniae type 22F capsular polysaccharide diphtheria CRM197 protein conjugate antigen is a component of modern pneumococcal conjugate vaccines such as Prevnar 20 and Vaxneuvance. It consists of the purified capsular polysaccharide from S. pneumoniae serotype 22F chemically linked (conjugated) to the non-toxic mutant diphtheria toxin carrier protein CRM197. This glycoconjugation enhances immunogenicity by inducing a T-cell dependent immune response, leading to robust antibody production and immunological memory even in young children. The primary mechanism of action is the induction of anti-capsular antibodies that target S. pneumoniae serotype 22F, facilitating opsonization and clearance by phagocytes. The inclusion of this serotype in multivalent vaccines broadens protection against invasive pneumococcal diseases (IPD), including bacteremia and meningitis, as well as non-invasive diseases like otitis media and pneumonia caused by S. pneumoniae[2][3][5]. These vaccines are indicated for active immunization to prevent disease caused by included serotypes in both pediatric and adult populations.
