Streptococcus pneumoniae type 8 capsular polysaccharide diphtheria CRM197 protein conjugate antigen is a semisynthetic glycoconjugate vaccine candidate designed to protect against invasive disease caused by Streptococcus pneumoniae serotype 8, a highly virulent strain associated with frequent outbreaks and increasing antibiotic resistance. The vaccine consists of purified or synthetic tetrasaccharides from the serotype 8 capsule, chemically linked (conjugated) to the non-toxic mutant diphtheria toxin carrier protein CRM197. This conjugation enhances immunogenicity by inducing a T-cell dependent immune response, leading to robust antibody production and long-term protection. While serotype 8 is included in some existing pneumococcal vaccines as a plain polysaccharide (such as Pneumovax 23), it is not present in widely used conjugate vaccines like Prevnar13; thus, this candidate addresses an unmet need for broader coverage. The primary indication is prevention of invasive pneumococcal diseases such as bacteremia and pneumonia caused by S. pneumoniae serotype 8[1][5][7].
