STS-201 is an investigational small molecule drug candidate being developed by Rize Oncology for the treatment of soft tissue sarcoma and various solid tumors. Its mechanism of action is characterized by blocking the cell cycle at the late G2/M boundary, a phase where cells are particularly susceptible to radiation-induced damage. Beyond its primary role as a radiosensitizer, STS-201 promotes the normalization of tumor neo-vasculature. This normalization improves tumor oxygenation—further enhancing the efficacy of radiotherapy through the formation of reactive oxygen species—and optimizes the delivery of concurrent therapies. Additionally, the stabilization of the tumor's vascular structure is intended to reduce the drug's metastatic potential. STS-201 is being positioned as a potential neoadjuvant therapy and may qualify for Orphan Drug status for soft tissue sarcoma.
