STATegics' second-generation thrombopoietin (TPO) allomimetics, represented by the lead compound STS-T4, are orally available small molecules designed to treat thrombocytopenia and bone marrow malignancies. Unlike first-generation mimetics or endogenous TPO, these compounds function as allosteric modulators by binding to a physically distinct site on the thrombopoietin receptor (MPL). This mechanism allows them to induce the proliferation of human hematopoietic progenitor cells and their differentiation into megakaryocytes, either independently or in synergy with endogenous thrombopoietin. Preclinical studies have demonstrated promising potency and safety profiles for the treatment of low platelet counts and potentially leukemia.
