Sturlimogene erparepvec (RP2) is an engineered, next-generation oncolytic herpes simplex virus type 1 (HSV-1) developed by Replimune. It is derived from the same backbone as RP1 (vusolimogene erparepvec) but is further modified to express a monoclonal antibody-like molecule targeting CTLA-4 (anti-CTLA-4), in addition to a fusogenic protein (GALV-GP R-) and human GM-CSF. The virus is designed to selectively replicate within and lyse tumor cells, releasing tumor-associated antigens while simultaneously promoting a systemic anti-tumor immune response. The expression of the anti-CTLA-4 antibody directly into the tumor microenvironment is intended to block inhibitory immune checkpoints locally, potentially increasing efficacy while minimizing systemic toxicity compared to systemic checkpoint inhibitors. It is primarily administered via intratumoral injection.
