STX-0119 is a **selective small molecule inhibitor of signal transducer and activator of transcription 3 (STAT3)** dimerization, binding to the STAT3 SH2 domain and preventing its phosphorylation and subsequent transcriptional activity[3][8]. It downregulates the expression of several STAT3 target genes, including c-myc, Bcl-xL, survivin, cyclin D1, MMP9, TGF-β1, and VEGF, and has been shown to induce apoptosis in cancer and stem cell models[1]. Preclinical studies demonstrate efficacy in models of **glioblastoma, lymphoma, osteoarthritis, and fibrosis (kidney and liver)**, as well as modulation of immune cell infiltration and the tumor microenvironment[1][2][5][7]. STX-0119 has also shown potential to suppress inflammation and cartilage degeneration in osteoarthritis, via STAT3/PPARγ signaling[5]. The compound is research-stage and not approved for clinical use.
