STX-0640 is a preclinical-stage Cytotoxicity Targeting Chimera (CyTAC) being developed by Solu Therapeutics for the treatment of systemic mastocytosis and other immunodermatologic indications. The CyTAC platform utilizes a modular, bifunctional design consisting of a small molecule binder linked to a proprietary effector antibody. In the case of STX-0640, the small molecule component selectively binds Mas-related G-protein coupled receptor member X2 (MRGPRX2), a G-protein coupled receptor highly and selectively expressed on the surface of mast cells. The effector antibody component is a CD3-mediated T-cell engager. By simultaneously binding MRGPRX2 on mast cells and CD3 on T cells, STX-0640 recruits and activates T cells to selectively deplete pathogenic mast cells, which are key drivers of immunological and inflammatory diseases.
