SU056 is a **small molecule inhibitor** of Y box binding protein 1 (YBX1, also referred to as YB-1), a multifunctional protein implicated in tumor progression and emergence of treatment resistance. SU056 disrupts YB-1 function by inhibiting its binding to mRNA and its interaction with the protein synthesis machinery, leading to suppression of translation and tumor growth. SU056 shows potent anti-tumor activity in preclinical models of ovarian cancer and triple-negative breast cancer, inhibiting cell proliferation, drug resistance, and metastasis. It leads to cell cycle arrest (G1 phase), enhances apoptosis, and reduces multidrug resistance. SU056 has shown strong synergy when combined with paclitaxel in vivo. It exhibits good tolerability, with toxicological studies in animals demonstrating no observable adverse effects even at high doses. SU056 is being advanced toward clinical development by Oregon Health & Science University, with a startup company leading translational and enabling studies for future human trials[1][2][3][5][7].
