Drug intelligence / Profile preview
SUBA itraconazole is a novel oral formulation of the antifungal agent itraconazole that utilizes "super bioavailability" (SUBA) technology. This technology disperses the active drug in a pH-dependent polymeric matrix to enhance dissolution and intestinal absorption compared to conventional crystalline formulations. The result is improved and more consistent bioavailability with less interpatient variability and reduced food or acid effect on absorption. Mechanistically, like other triazole antifungals, it inhibits fungal cytochrome P450 14α-demethylase (lanosterol 14α-demethylase), thereby blocking ergosterol synthesis essential for fungal cell membrane integrity. It is FDA-approved for the treatment of systemic mycoses including blastomycosis, histoplasmosis (including chronic cavitary pulmonary disease and disseminated non-meningeal forms), and aspergillosis in patients intolerant of or refractory to amphotericin B therapy. It is also used off-label for dermatophytosis and onychomycosis[2][3][5][7][8].
