Overview

This subcutaneous allergen immunotherapy (SCIT) vaccine, developed by Inmunotek S.L., is a depot formulation designed for the treatment of allergic rhinitis, allergic rhinoconjunctivitis, and allergic asthma. The vaccine consists of a mixture of allergen extracts from grasses (typically *Phleum pratense*) and Cupressaceae (typically *Cupressus arizonica*), which are adsorbed onto aluminum hydroxide to provide a sustained release effect. The mechanism of action involves the induction of immunological tolerance through repeated exposure to specific allergens. This process shifts the immune response from a Th2-dominated profile, associated with IgE production and allergic inflammation, to a Th1/Treg-dominated profile. This shift is characterized by the production of allergen-specific IgG4 "blocking" antibodies and regulatory cytokines such as IL-10 and TGF-beta, which ultimately reduce the activation and recruitment of mast cells, basophils, and eosinophils upon subsequent allergen exposure.
