Subtype C gp140 is a recombinant protein subunit HIV vaccine candidate developed by Novartis in collaboration with the HIV Vaccine Trials Network (HVTN) and the South African AIDS Vaccine Initiative (SAAVI). It is based on the envelope glycoprotein of HIV-1 Subtype C, specifically the TV1 strain, which is highly prevalent in Southern Africa. The gp140 protein is a soluble, truncated version of the viral envelope (Env) protein, encompassing the gp120 and the extracellular domain of gp41, designed to expose key epitopes for the induction of neutralizing antibodies. In clinical trials such as HVTN 086, it is typically administered with the MF59 adjuvant as part of a heterologous prime-boost regimen involving DNA (SAAVI DNA-C2) and MVA (SAAVI MVA-C) vectors to enhance the breadth and magnitude of the immune response against HIV-1.
