Overview

Succinylsulfathiazole is a synthetic sulfonamide antibacterial agent primarily used for its local action in the gastrointestinal tract. It is classified as an ultra long-acting prodrug of sulfathiazole and is poorly absorbed from the gut; about 95% remains in the intestine where it exerts its effect. The drug acts by competitively inhibiting bacterial dihydropteroate synthetase, thereby blocking folic acid synthesis in bacteria. This results in suppression of folate-producing gut flora and prevention or treatment of gastrointestinal infections. Succinylsulfathiazole was historically used to reduce bacterial load before bowel surgery and to treat intestinal infections such as bacillary dysentery. It has also been employed experimentally to induce folate deficiency for research purposes[4][5][6][7]. The drug is slowly hydrolyzed by intestinal bacteria to release active sulfathiazole[4][6].
