Overview

Sugemalimab + radiation refers to the combination of **sugemalimab**, a fully human, full-length anti–PD-L1 IgG4 monoclonal antibody, with **radiation therapy** (including potential use of low-dose radiotherapy). Sugemalimab blocks PD-L1, restoring T-cell activity to enhance anti-tumor immunity. This combination is under study for synergistic anti-cancer effects, where radiation may increase immunogenicity of the tumor microenvironment, potentially enhancing response to immunotherapy. Principal indications under investigation include non-small cell lung cancer (NSCLC), specifically in extensive-stage small cell lung cancer (ES-SCLC) with positive SLFN-11 biomarkers, where both are being combined with chemotherapy and maintenance therapies in clinical trials. Sugemalimab was developed using the OmniRat transgenic animal platform, reducing the risk of immunogenicity. Radiation therapy is a standard local therapy for cancer, causing tumor cell death primarily through DNA damage.
