Overview

SUL-138 is a small molecule 6-chromanol derivative inspired by the metabolic plasticity of hibernating animals. It functions as a mitochondrial bioenergetics enhancer by supporting the activity of Complex I (NADH dehydrogenase) and Complex IV (cytochrome c oxidase) within the electron transport chain. This mechanism promotes ATP production and reduces the generation of reactive oxygen species (ROS) without affecting basal mitochondrial membrane potential. Preclinical studies in Alzheimer's disease models demonstrate that SUL-138 is blood-brain barrier permeable, rescues synaptic plasticity and memory deficits, reduces amyloid-beta plaque load, and shifts mitochondrial metabolism toward fatty acid oxidation. It has also shown potential in treating renal ischemia/reperfusion injury, chronic obstructive pulmonary disease (COPD), and diabetic kidney damage.
