Overview

**sulfadiazine + sulfamethazine + sulfamerazine** is a fixed-dose combination of three sulfonamide antibiotics, historically referred to as "triple sulfa" or "trisulfapyrimidines". Each component inhibits the enzyme dihydropteroate synthetase in bacteria, competitively blocking the incorporation of para-aminobenzoic acid (PABA) into dihydropteroate, thereby preventing the synthesis of dihydrofolic acid and subsequent folate-dependent DNA processes. This results in bacteriostatic effects against a broad range of Gram-positive and Gram-negative bacteria. Triple sulfa was developed to provide synergistic antibacterial coverage and to reduce the likelihood of resistance. The combination has been used orally and topically, particularly for the treatment of bacterial infections, such as urinary tract infections, streptococcal pharyngitis, and conjunctivitis, as well as for prophylactic applications.
