Overview
Sulfadoxine is a long-acting sulfonamide antibiotic primarily used as an antimalarial agent, almost exclusively in fixed-dose combination with pyrimethamine. It acts by inhibiting the enzyme dihydropteroate synthase, which blocks the conversion of para-aminobenzoic acid (PABA) to folic acid—a process essential for DNA synthesis and cell growth in susceptible organisms such as Plasmodium falciparum. By interfering with folate synthesis, sulfadoxine impairs parasite replication and survival. Sulfadoxine has also been used to treat certain bacterial infections in veterinary medicine[1][8].
