sulfadoxine + pyrimethamine

Sulfadoxine + pyrimethamine is an oral fixed-dose antimalarial combination comprising the long-acting sulfonamide sulfadoxine and the antifolate pyrimethamine. Sulfadoxine competitively inhibits *Plasmodium* dihydropteroate synthase and pyrimethamine inhibits *Plasmodium* dihydrofolate reductase, producing sequential blockade of parasite folate synthesis and synergistic inhibition of nucleic-acid synthesis. It is used for treatment and prevention of malaria, including intermittent preventive treatment in pregnancy and, with amodiaquine, seasonal malaria chemoprevention in eligible endemic-area populations; resistance associated with parasite *dhps* and *dhfr* mutations limits effectiveness in many settings.
