Overview

This is a six-drug antimalarial combination consisting of sulfadoxine, pyrimethamine, chloroquine, amodiaquine, primaquine, and artesunate. Each component targets different stages or mechanisms in the malaria parasite's lifecycle: - **Sulfadoxine** is a long-acting sulfonamide that inhibits dihydropteroate synthase in the folate synthesis pathway. - **Pyrimethamine** is an antiprotozoal that inhibits protozoal dihydrofolate reductase. - **Chloroquine** and **amodiaquine** are 4-aminoquinoline compounds that interfere with heme detoxification in the parasite’s food vacuole. - **Primaquine** is an 8-aminoquinoline effective against liver (hypnozoite) stages of Plasmodium vivax and Plasmodium ovale. - **Artesunate** is an artemisinin derivative that generates free radicals to damage parasitic proteins. This combination would theoretically provide broad-spectrum activity against multiple Plasmodium species (including resistant strains), targeting both blood and liver stages. However, there are no known marketed products or clinical guidelines recommending this exact six-drug combination as a single therapy; rather, various subsets (such as sulfadoxine-pyrimethamine plus artesunate or amodiaquine) are used for specific indications[2][3][7]. The use of all six together would be highly unusual due to overlapping toxicities and drug interactions.
