Overview

A combination of the antimalarial drugs sulfadoxine and pyrimethamine with folic acid supplementation. Sulfadoxine inhibits dihydropteroate synthase while pyrimethamine inhibits dihydrofolate reductase, both interfering with folate metabolism in the malaria parasite. Folic acid is added to prevent neural tube defects in pregnant women while maintaining antimalarial efficacy. The efficacy of sulfadoxine-pyrimethamine can be reduced by high doses of folic acid (≥5 mg), but lower doses (0.4-0.8 mg) do not significantly affect treatment outcomes.
