Sulfaphenazole is a sulfonamide antibacterial drug that acts as a competitive inhibitor of the bacterial enzyme dihydropteroate synthase (DHPS), which is involved in folate synthesis. By inhibiting this enzyme, sulfaphenazole disrupts folate production in bacteria and exerts bacteriostatic effects[2][7]. In addition to its antibacterial activity, sulfaphenazole is a highly selective inhibitor of the human cytochrome P450 isozyme CYP2C9 (Ki ≈ 0.3 μM), with much lower activity against other CYP450 isoforms[1][5]. It has been used clinically under brand names such as Sulfabid for the treatment of bacterial infections and has also been studied for its effects on vascular tone via inhibition of endothelium-derived hyperpolarizing factor synthase (a CYP450 isozyme)[3][5].
