Sulofenur is a synthetic diarylsulfonylurea classified as an antineoplastic agent, developed for its broad spectrum of activity against various solid tumors in preclinical models[1][4][7]. Its precise mechanism of antitumor action remains unresolved, though evidence suggests it inhibits ribonucleotide reductases, which are crucial enzymes for DNA synthesis[7]. Sulofenur underwent extensive clinical investigations (phase I and II) as an oral small molecule for advanced cancers, but development was ultimately discontinued due to dose-limiting toxicities including hemolytic anemia and methemoglobinemia, and lack of significant efficacy in trials[3][4][6][7].
