Sumanirole is a highly selective dopamine D2 receptor full agonist developed primarily for the treatment of Parkinson's disease and restless legs syndrome. It was the first compound discovered with such high selectivity for the D2 receptor over other dopamine receptor subtypes (notably D1, D3, and D4). Sumanirole acts by stimulating dopamine D2 receptors in the brain, which are implicated in motor control and are deficient in Parkinson's disease due to degeneration of dopaminergic neurons. The drug demonstrated anti-parkinsonian activity in preclinical models and was also investigated for use in restless legs syndrome. Despite promising pharmacological properties—including >200-fold selectivity for D2 over other subtypes—clinical development was discontinued after studies failed to show sufficient benefit over existing therapies. Sumanirole remains a valuable research tool compound[1][5][3][6].
