Sunitinib + bevacizumab + carboplatin + paclitaxel is an investigational combination therapy that was evaluated for the first-line treatment of locally advanced, metastatic, or recurrent nonsquamous non-small cell lung cancer (NSCLC). The regimen consists of **sunitinib** (a small molecule multi-targeted receptor tyrosine kinase inhibitor), **bevacizumab** (a monoclonal antibody targeting VEGF), and the chemotherapy doublet **carboplatin** and **paclitaxel**. The combination was studied in the Phase II **SABRE-L** trial (Study Assessing the Blockade of both VEGF Receptor and ligand to enhance Efficacy in Lung) with the rationale of achieving dual blockade of the VEGF pathway (targeting both the ligand and its receptors) to improve clinical outcomes. However, clinical results indicated that the addition of sunitinib to the standard paclitaxel/carboplatin/bevacizumab regimen was poorly tolerated at the tested doses, primarily due to severe myelosuppressive toxicities. Consequently, the development of this specific four-drug combination was terminated.
