Sunitinib + chlorin e6 (specifically formulated as SCF NPs) is an experimental multifunctional theranostic nanoplatform designed for the synergistic treatment of hepatocellular carcinoma (HCC). This system integrates sunitinib, a multi-targeted tyrosine kinase inhibitor (TKI), with chlorin e6 (Ce6), a second-generation photosensitizer, within a folic acid-functionalized PEGylated polymeric framework (Ce6-PEG2000-FA). Sunitinib provides molecular targeted therapy by inhibiting multiple receptor tyrosine kinases, including VEGFR and PDGFR, which suppresses tumor angiogenesis and growth. Chlorin e6 enables both photodynamic therapy (PDT) and photothermal therapy (PTT) upon laser irradiation, generating reactive oxygen species and heat to induce localized tumor cell death. The folic acid component facilitates active targeting to folate receptors, which are frequently overexpressed in HCC. Preclinical studies have demonstrated that this combination offers multimode imaging capabilities and superior tumor inhibition compared to monotherapy, potentially leading to complete tumor eradication in animal models.
