Overview

Sunitinib + FOLFIRI is an investigational combination therapy used in clinical trials for the treatment of metastatic colorectal cancer (mCRC). Sunitinib is a small molecule multi-targeted tyrosine kinase inhibitor that blocks several receptors involved in tumor angiogenesis and proliferation, including vascular endothelial growth factor receptors (VEGFRs), platelet-derived growth factor receptors (PDGFRs), KIT, FLT3, RET, and CSF1R. By inhibiting these kinases, sunitinib disrupts tumor blood supply and cell signaling pathways essential for cancer growth. FOLFIRI is a standard chemotherapy regimen composed of three drugs: folinic acid (leucovorin), which enhances the efficacy of fluorouracil; fluorouracil (5-FU), an antimetabolite that inhibits DNA synthesis; and irinotecan, a topoisomerase I inhibitor that prevents DNA replication. The combination aims to leverage both targeted antiangiogenic effects from sunitinib with cytotoxic effects from chemotherapy to improve outcomes in mCRC patients[1][2][3][4].
