Sup19 CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy targeting the B-lymphocyte antigen CD19. Developed by the Institute of Hematology & Blood Diseases Hospital at the Chinese Academy of Medical Sciences, this therapy is specifically engineered to address clinical needs in patients with relapsed or refractory B-cell malignancies who have either failed prior CD19-targeted therapies or whose tumors exhibit low/weak CD19 expression. The "Sup19" construct is designed to provide enhanced sensitivity to low-density antigens, potentially overcoming common resistance mechanisms such as antigen escape or downregulation that lead to relapse after standard CAR-T treatments. It is currently being evaluated in Phase 1 clinical trials for B-cell acute lymphoblastic leukemia (B-ALL) and B-cell lymphoblastic lymphoma (B-LBL).
