Super-GDF9 is a bioengineered, high-potency variant of human Growth Differentiation Factor 9 (GDF9) designed to improve oocyte developmental competence in assisted reproductive technologies. It was created by incorporating four specific amino acid residues from Bone Morphogenetic Protein 15 (BMP15)—Arg301, Gly304, His307, and Met369—into the human GDF9 sequence. These modifications allow Super-GDF9 to bypass the natural latency of wild-type GDF9, enabling it to signal potently as a homodimer without the requirement for heterodimerization with BMP15 (the natural "cumulin" complex). Super-GDF9 acts as a potent agonist for the SMAD2/3 signaling pathway via the type I receptors Activin receptor-like kinase 4 (ALK4) and 5 (ALK5). It is primarily being investigated as a supplement for Capacitation-In Vitro Maturation (CAPA-IVM) culture media to enhance cumulus cell expansion and improve embryo quality, particularly for patients with Polycystic Ovary Syndrome (PCOS) or those at high risk for Ovarian Hyperstimulation Syndrome (OHSS).
