Drug intelligence / Profile preview
Super-interferon gamma (also known as super-human IFNγ or super-IFNγ) is an engineered, mutant form of human interferon-gamma (IFNγ) designed using computational tools (such as FoldX and ModelX) to introduce point mutations that enhance thermal stability, production, and receptor binding. Developed by the Center for Genomic Regulation (CRG) and the Hospital del Mar Research Institute (IMIM), this cytokine variant exhibits a 10-fold increase in biological activity and a 7-fold improvement in production compared to wild-type IFNγ. To overcome the limitations of recombinant IFNγ, such as high aggregation and a short half-life, super-interferon gamma is delivered locally and intratumorally using an attenuated bacterial chassis. In preclinical rodent models of breast cancer, intratumoral delivery of super-interferon gamma significantly enhances immune cell recruitment, boosts inflammatory gene signatures within the tumor microenvironment, and exerts strong anti-proliferative and anti-tumor effects.
