SuperMApo is an investigational biological drug candidate composed of the supernatant of apoptotic leukocytes. Developed by the Établissement Français du Sang (EFS), it is designed to exploit the anti-inflammatory and pro-resolving signaling pathways associated with efferocytosis, the process by which macrophages clear apoptotic cells. SuperMApo contains a complex array of factors, including Transforming Growth Factor-beta 1 (TGF-β1), which act on myeloid cells to induce a regulatory macrophage (Mreg) phenotype. This transition results in the increased secretion of Interleukin-10 (IL-10) and the inhibition of pro-inflammatory cytokines such as Tumor Necrosis Factor-alpha (TNF-α) and Interleukin-6 (IL-6). The therapy is primarily being developed for the treatment of chronic Graft-versus-Host Disease (cGvHD) and other autoimmune or inflammatory conditions to restore immune homeostasis and promote tolerance.
