Supervax is a prophylactic subunit vaccine for the prevention of hepatitis B virus (HBV) infection. It is composed of recombinant hepatitis B surface antigens (HBsAg) and the synthetic adjuvant RC-529, which is a monosaccharide analog of lipid A that functions as a Toll-like receptor 4 (TLR4) agonist. Unlike other lipid A-based adjuvants like MPL or GLA which have disaccharide backbones, RC-529 features a monosaccharide backbone. The vaccine was licensed in Argentina in 2003 and is administered as a two-dose regimen. Clinical data indicates that the RC-529-adjuvanted formulation provides significantly higher seroprotection rates and geometric mean anti-HBs titers compared to traditional aluminum-adjuvanted hepatitis B vaccines.
