Overview

Supinoxin (RX-5902) is an orally administered small molecule and first-in-class inhibitor of phosphorylated-p68 RNA helicase (P-p68/DDX5), a member of the DEAD box family of RNA helicases. By inhibiting P-p68 phosphorylation and function, supinoxin blocks nuclear translocation of β-catenin and disrupts Wnt/β-catenin signaling pathways implicated in cell proliferation and cancer progression. Preclinical studies have shown potent antiproliferative activity against triple-negative breast cancer (TNBC) cell lines as well as efficacy in animal models for various solid tumors including breast cancer. Clinical development focused on advanced or metastatic solid tumors—especially TNBC—but has been discontinued for all indications[1][2][4][5][6].
