Overview

SUPLEXA is a personalized autologous cellular immunotherapy developed by Alloplex Biotherapeutics for the treatment of advanced cancers. It is manufactured from a patient’s own peripheral blood mononuclear cells (PBMCs), which are reprogrammed ex vivo using the proprietary ENLIST cell platform. This process activates and expands multiple immune effector cell types—including natural killer (NK) cells, natural killer T (NKT) cells, γδ T cells, and cytotoxic CD8+ and CD4+ αβ T lymphocytes—without genetic engineering. The resulting product harnesses both innate and adaptive immune mechanisms to recognize and kill tumor cells through multiple pathways. Early clinical trials have demonstrated promising safety and efficacy signals in patients with solid tumors (notably colorectal cancer and renal cell carcinoma) as wellalike hematologic malignancies such as lymphoma and chronic lymphocytic leukemia[1][2][3][4][5][6][9].
