Overview

Suprofen is a nonsteroidal anti-inflammatory drug (NSAID) of the propionic acid derivative class, structurally related to ibuprofen, ketoprofen, and naproxen. It was developed by Janssen Pharmaceutica and marketed primarily as a 1% ophthalmic solution under the brand name Profenal. Suprofen acts as an analgesic, antipyretic, and anti-inflammatory agent. Its primary mechanism of action is inhibition of cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2), leading to reduced synthesis of prostaglandins from arachidonic acid and thus decreasing inflammation and pain. Clinically, it was used to prevent intraoperative miosis during ocular surgery but has been discontinued in the United States due to safety concerns regarding renal effects when used systemically[1][2][3][6].
