Drug intelligence / Profile preview
Suregraft (everolimus) is a branded generic formulation of everolimus, an orally bioavailable inhibitor of the mechanistic target of rapamycin (mTOR) pathway. Developed by Hospimax, it is primarily indicated for the prevention of organ rejection in patients receiving allogeneic renal, cardiac, or hepatic transplants, typically in combination with other immunosuppressive agents such as cyclosporine or tacrolimus and corticosteroids. Everolimus acts by binding to the intracellular receptor protein FKBP-12 (FK506-binding protein 12) to form an active complex that subsequently binds to and inhibits the mTOR complex 1 (mTORC1). This inhibition blocks downstream signaling pathways, including the phosphorylation of p70 S6 kinase, thereby suppressing interleukin-stimulated T- and B-lymphocyte proliferation and clonal expansion. In addition to its immunosuppressive properties, everolimus is also utilized in oncology to treat various malignancies, including advanced renal cell carcinoma, hormone receptor-positive breast cancer, and pancreatic neuroendocrine tumors.
