Overview

Surinabant (SR147778) is an orally active, selective cannabinoid receptor type 1 (CB1) antagonist developed by Sanofi-Aventis. It was investigated primarily as a potential treatment for nicotine addiction and smoking cessation, with additional research into its use for obesity, alcoholism, and other addictive disorders. Surinabant acts by blocking the CB1 receptor, which plays a key role in regulating appetite and reward pathways. In preclinical models, it reduced body weight gain and improved metabolic parameters such as plasma glucose and triglyceride levels. Clinical trials reached up to phase II but were discontinued for all major indications including obesity, alcoholism, psychotic disorders, and smoking withdrawal due to limited efficacy or adverse effects at higher doses[2][3][4][5][7].
