Overview

The surrogate murine MERTK antibody is a preclinical monoclonal antibody developed by Rgenix to model the therapeutic activity of their clinical candidate, RGX-019, in murine systems. Because RGX-019 is highly specific for human MERTK and does not cross-react with the murine ortholog, this surrogate antibody was generated to evaluate the effects of MERTK inhibition within an intact murine immune system. The antibody functions by binding to the MERTK receptor, triggering its rapid internalization and degradation from the cell surface. This mechanism disrupts downstream AKT signaling, which is typically activated by the ligand Gas6 to promote tumor cell survival, proliferation, and migration. Furthermore, the antibody modulates the tumor microenvironment by shifting immunosuppressive M2 macrophages toward a pro-inflammatory phenotype. Preclinical studies have demonstrated that this surrogate antibody can significantly inhibit the growth of breast cancer tumors in vivo.
