Surufatinib + envafolimab is an investigational combination therapy consisting of two agents with complementary mechanisms of action for the treatment of advanced solid tumors, including neuroendocrine neoplasms and biliary tract carcinoma. Surufatinib is an oral small-molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors 1, 2, and 3 (VEGFR1/2/3), fibroblast growth factor receptor 1 (FGFR1), and colony-stimulating factor 1 receptor (CSF-1R). This results in inhibition of tumor angiogenesis and modulation of the tumor immune microenvironment[4][7]. Envafolimab is a humanized single-domain anti-programmed cell death protein 1 (PD-1) monoclonal antibody administered subcutaneously; it blocks PD-1 to enhance T-cell mediated antitumor immunity[9][10]. The combination aims to leverage both antiangiogenic effects and immune checkpoint blockade for synergistic antitumor activity. Early-phase clinical trials are ongoing in indications such as unresectable or metastatic biliary tract cancer, soft tissue sarcoma, and other advanced solid tumors[7].
