Drug intelligence / Profile preview
Surufatinib + sintilimab + capecitabine is an investigational three-drug combination regimen. It is being evaluated in an investigator-initiated Phase Ib/II clinical trial (NCT05472948) for the treatment of previously treated metastatic small bowel adenocarcinoma (SBA) and appendiceal carcinoma. The regimen combines surufatinib, an oral small-molecule multi-kinase inhibitor targeting VEGFR1-3, FGFR1, and CSF-1R; sintilimab, a recombinant humanized IgG4 monoclonal antibody targeting PD-1; and capecitabine, an oral fluoropyrimidine carbamate chemotherapy that acts as a prodrug of 5-fluorouracil (5-FU). This combination aims to leverage the synergistic effects of anti-angiogenesis, CSF-1R-directed tumor microenvironment reprogramming, immune checkpoint blockade, and cytotoxic chemotherapy.
