Overview

This treatment regimen is a multi-modal combination therapy being evaluated for the treatment of unresectable locally advanced esophageal squamous cell carcinoma (ESCC). It consists of **surufatinib**, a small molecule angio-immuno kinase inhibitor that targets VEGFR1/2/3, FGFR1, and CSF1R; **toripalimab**, a monoclonal antibody that acts as a PD-1 checkpoint inhibitor; and a chemotherapy backbone of **albumin-bound paclitaxel** and **cisplatin**. The regimen is typically administered in a neoadjuvant setting, often followed by or concurrent with definitive thoracic intensity-modulated radiotherapy (IMRT). Surufatinib's mechanism of action involves inhibiting tumor angiogenesis and modulating the tumor immune microenvironment by reducing tumor-associated macrophages, which is intended to synergize with the T-cell activation provided by toripalimab and the cytotoxic effects of chemotherapy to improve resectability and overall prognosis.
