Overview

The survivin-2B peptide vaccine is a synthetic therapeutic cancer vaccine designed to induce an immune response against tumor cells expressing the survivin protein, specifically targeting the 2B splice variant of survivin (also known as Baculoviral IAP repeat-containing protein 5). Survivin is an inhibitor of apoptosis protein that is highly expressed in various cancers but minimally present in normal adult tissues. The vaccine consists of a short synthetic peptide corresponding to amino acids 80–88 (AYACNTSTL) of the human survivin-2B isoform and is typically administered with or without immunostimulatory adjuvants such as interferon-beta or Montanide ISA51VG. Its mechanism involves stimulating cytotoxic T lymphocytes (CTLs) specific for HLA-A24-restricted epitopes on tumor cells, leading to targeted cell lysis. Clinical trials have evaluated its safety and immunogenicity in advanced pancreatic cancer, gastrointestinal cancers, and oral squamous cell carcinoma. While generally safe and well-tolerated with minimal adverse effects reported (mainly mild injection site reactions), clinical efficacy has been limited when used alone; combination with adjuvants may enhance immune responses[1][3][4][5][6][8].
