A survivin-loaded dendritic cell is an **autologous cell therapy** in which dendritic cells (DCs), a type of immune cell, are loaded ex vivo with the tumor-associated antigen survivin. These cells are then administered back to the patient to stimulate an anti-tumor immune response. Survivin (also known as BIRC5) is a protein highly expressed in a variety of cancers but minimally in normal adult tissues, making it an attractive immunotherapy target. In practice, patient-derived monocyte DCs are exposed to survivin peptides or survivin mRNA, leading to the presentation of survivin-derived epitopes on their surface. Upon reinfusion, these DCs enhance T-cell mediated immune responses against survivin-expressing tumor cells. This investigational therapy has mainly been evaluated in clinical trials for melanoma, non-small cell lung cancer, and prostate cancer, often in combination with other antigens (e.g., hTERT, MAGE-3, PSMA) and/or adoptive T cell therapies. No product with this exact configuration is FDA/EMA approved as of 2025, and clinical activity remains investigational[1][2][3][4].
