**Survivin mRNA Galsome** is a preclinical therapeutic cancer vaccine for multiple myeloma developed using the Galsome platform. It consists of lipid nanoparticles co-encapsulating mRNA encoding survivin, also known as baculoviral IAP repeat-containing protein 5, and the invariant natural killer T-cell agonist alpha-galactosylceramide. The survivin mRNA is intended to drive antigen presentation and survivin-specific CD8-positive T-cell responses against myeloma cells, while alpha-galactosylceramide is presented by CD1d to activate invariant natural killer T cells. In the 5T33MM mouse model, prime-boost treatment reduced myeloma burden, but repeat vaccination was associated with loss of survivin-reactive T-cell populations, consistent with possible fratricide because survivin can be expressed by activated T cells. The program remains preclinical.
